Overview

Förster Resonance Energy Transfer is investigated in single molecule experiments.

Complementary DNA oligonucleotides, where one strand carries a donor dye, while the other carries an acceptor dye, are annealed and immobilized using microfluidics on a quartz surface.

With a Total Internal Reflection Fluorescence Microscope (TIRFM), the FRET-efficiency of single-molecule FRET events is measured and used to determine the donor-acceptor distance.
