Superresolution Fluorescence Microscopy

Overview

A single-molecule localization microscope (SMLM) is used to resolve structures below the classical resolution limit (‘sub-diffraction imaging’)

Specific template regions within DNA origami are localized by transient binding of labeled DNA oligonucleotides (DNA PAINT technique). With aid of these nanoruler positions, the localization precision of the instrument is characterized.

The actin network within HeLa cells is studied with the dSTORM technique and compared to  results from conventional fluorescence imaging.
