Fluorescence Lifetime

Idea: In this experiment the lifetime of fluorescent molecules is used to determine
internal dynamics and distances in simple molecular systems

Overview

Time-correlated single photon counting is used to determine fluorescence lifetimes.

The influence of intramolecular bridges and attachment to DNA on fluorescence lifetime of a fluorophore is investigated.

The distance between two fluorophores attached to complementary DNA strands is determined from the decrease of the donor lifetime in presence of the acceptor due to Förster Resonance Energy Transfer (FRET).
